Sure, you can. But by whisking in air, you give the oil something to form bubbles around, and you'll get a better emulsion.

After a recent thread on making a vinaigrette, I made the emulsion by shaking the ingredients vigorously in a canning jar - one of those Ball or Mason jars. The results were surprising. I made the vinaigrette almost two weeks ago, and it still hasn't separated. I am quite happy with the result.
